Magnesium's role in sleep has attracted significant research attention — and the evidence is growing. Magnesium not only reduces the excitability of the nervous system and alters muscle relaxation but also regulates cellular biological clocks, energy balance and circadian rhythms, playing a crucial role in sleep regulation.
A landmark 2025 RCT published in Nature & Science of Sleep by Schuster et al. enrolled 155 adults aged 18–65 years with self-reported poor sleep quality and found magnesium bisglycinate supplementation significantly improved sleep vs placebo. Taking magnesium L-threonate improved both subjective and objective sleep scores and daytime functioning.

Calcium triggers muscle contraction. Magnesium promotes relaxation by competing with calcium at receptor sites on muscle cell membranes. 🔬 Research AssociationBlood Sugar Regulation
Magnesium is a cofactor for insulin receptor signalling — meaning it plays a role in how efficiently cells respond to insulin. Low magnesium is consistently associated with insulin resistance and type 2 diabetes in observational studies. A pooled analysis of 24 RCTs published in Frontiers in Nutrition (Xu et al., 2023) found magnesium supplementation significantly improved markers of hyperglycaemia and hypercholesterolaemia in type 2 diabetes patients.
There is also a bidirectional relationship — type 2 diabetes increases urinary magnesium excretion, further depleting levels. This feedback loop makes magnesium status particularly important for those managing blood sugar.

This is one of the most important and most overlooked roles of magnesium. Vitamin D3 from sunlight or supplements is biologically inactive — it must be converted into its active form (calcitriol) through two hydroxylation steps, both of which require magnesium as a cofactor. Without adequate magnesium, Vitamin D cannot be properly activated — meaning Vitamin D blood levels may appear adequate while the active hormone remains impaired.
Research by Deng et al. (2013) established this mechanism clearly. The practical implication: if you supplement Vitamin D, ensuring adequate magnesium intake is essential for the Vitamin D to work properly.

Emerging research is examining magnesium's relationship with inflammation and oxidative stress. A systematic review and meta-analysis aimed to evaluate the antioxidant effects of dietary and supplemental magnesium on several biomarkers related to oxidative stress and inflammation. A systematic search of studies published from 2000 to 2025 identified 28 relevant articles, including both animal and human studies. The review found associations between magnesium intake and markers of oxidative stress and inflammation.
Low magnesium is associated with elevated CRP (C-reactive protein) — a marker of systemic inflammation. Whether supplementation directly reduces inflammation in otherwise healthy adults remains an active area of research.
