1. What NMN actually is

NMN — β-Nicotinamide Mononucleotide, CAS 1094-61-7 — is a nucleotide found naturally in small quantities in food. In human biochemistry it sits one step upstream of NAD+ (nicotinamide adenine dinucleotide) in the salvage pathway. That is a description of where the molecule sits in a metabolic pathway. It is not a statement that taking it will do anything for you.

This distinction matters more than it might sound. NMN has no authorised health claims on the GB Nutrition and Health Claims Register. No UK seller — us included — is permitted to tell you that NMN improves energy, slows ageing, repairs anything, or produces any effect in the body. If you see a UK NMN listing making those claims, the claim is not authorised, whatever the science may or may not eventually show.

Human research does exist and is worth reading in the original. The first placebo-controlled human trial of oral NMN was published by Irie and colleagues in Endocrine Journal (2020), a small single-dose safety study in ten men. Yoshino and colleagues, writing in Science (2021), ran a 10-week randomised trial in 25 postmenopausal women with prediabetes. Later work by Liao and colleagues (JISSN, 2021) looked at amateur runners, and Yi and colleagues (GeroScience, 2023) at physical performance measures. These are small, short studies with mixed and sometimes modest findings. Read them and draw your own conclusion — that is a more honest position than any supplement seller summarising them for you.

2. The buyer's checklist — eight verifiable points

Every item here can be checked before you pay. None of them requires trusting the seller.

1 · A purity percentage, not the word "pure"

"High purity" is marketing. "99.99% by HPLC" is a specification. If no number is published, ask for one before you buy.

2 · A named laboratory — and what it tested

"Third-party tested" without a lab name is unverifiable. Then ask the question most buyers never think to ask: was the certificate run on the raw powder or on the finished capsule? A powder certificate issued before encapsulation tells you about the powder, not about what ended up in the shell.

3 · The capsule size, declared

Capsule shells have fixed volumes. A size 0 shell holds roughly 500–530mg of NMN powder; size 1 and size 2 hold materially less. If a 500mg declaration appears alongside a smaller shell, ask how.

4 · The filled capsule weight

5 · The ingredient list, read properly

Ratings are public at ratings.food.gov.uk. Check which premises the rating covers — a rating on an office or a distribution unit is not a rating on a food production facility.

8 · A trading history you can look up

3. Why it matters who fills the capsule

Most buyers compare purity percentage and price. Neither tells you who handled the powder. The typical supplement supply chain looks like this:

The certificate of analysis is normally issued at step one, on the powder. The brand at step three usually never sees the inside of step four. Nobody tests the finished capsule. This is the industry norm, not a scandal — but it does explain why a powder certificate is not the same as a finished-product certificate.

4. Four checks you can run at home

Read this before you run them. These are indicators, not analysis. Powder density, moisture content and normal fill tolerance all move the numbers. None of these checks identifies a substance or proves what a capsule contains — only a laboratory can do that. Treat an unexpected result as a question to put to the seller, not as a finding.

Check 1 — Capsule size

Compare the shell against a known size 0. Shell volumes are fixed and published, so this is the one physical check with a hard answer: a smaller shell cannot hold as much powder as a larger one. What it cannot tell you is what the powder is.

Check 2 — Weight

Weigh five capsules on a 0.01g scale and average. Roughly 0.60g for a 500mg size 0, roughly 0.82g for a 700mg size 00. A consistent, substantial shortfall against the declared fill is worth raising with the supplier. A few tens of milligrams either way is normal manufacturing variance.

Check 3 — The sublingual test (the one we recommend)

This is the check we point customers to, ahead of any of the others. Open a capsule and place a small amount of the contents under your tongue — the shell plays no part, so set it aside.

Check 4 — Read the ingredient list again

The most reliable check is also the least dramatic. Ingredients are listed in descending order by weight, so the order tells you something the front label will not. If a capsule declares NMN and the HPMC shell and nothing else, that is a complete answer. If it declares more, those ingredients are there for a reason — some are bulking agents, some are processing aids, some genuinely modify how the product releases. Ask which, rather than guessing.

Why we no longer suggest the glass-of-water test. We used to. It sounds decisive and it isn't: several perfectly legitimate excipients are simply insoluble in cold water, and hydrophilic matrix ingredients such as hypromellose are specifically designed to hydrate and slow release.
