Why NMN Dosage Matters More Than Most People Realise
NMN raises NAD+ levels. NAD+ declines with age. The relationship between age and NAD+ decline is well established — and it is the fundamental reason why dosage cannot be treated as a one-size-fits-all number.
NAD+ levels decline by roughly 50% between your 20s and 50s. For adults under 40, NAD+ decline is less pronounced and a 250mg daily dose is a reasonable entry point. Under 40 years: consider 250mg daily for maintenance — NAD+ decline is less pronounced at this age and moderate doses provide solid benefits.
The landmark Yoshino et al. (2021) study in Cell Metabolism used 250mg daily and showed improved skeletal muscle insulin sensitivity in postmenopausal women — demonstrating that 250mg can produce measurable metabolic effects in the right population. However, for younger adults without significant NAD+ depletion, 250mg is appropriate.
For anti-ageing purposes, 500mg per day is a well-supported starting dose for adults in their 40s and 50s. The GeroScience multicenter trial (2022) tested 300mg, 600mg and 900mg daily in 80 middle-aged adults (40–65 years) over 60 days and found all doses significantly increased blood NAD+ levels — with 600mg producing optimal NAD+ elevation.
The RENEW Study (2025) using 500mg NMN for 60 days in adults with a mean age of 52 found a 44% increase in intracellular NAD+ levels — importantly measuring intracellular rather than blood NAD+, which is more relevant to cellular function.
Adults over 60 may benefit from 500–900mg daily, based on clinical trials showing enhanced physical function at higher doses. This is where underdosing is most common — and most consequential. Adults over 50 taking 250mg are very likely not providing sufficient NMN to meaningfully restore NAD+ levels given the steeper decline at this age.
A January 2026 randomised controlled trial in 65 healthy adults found that 1,000mg per day NMN doubled circulating NAD+ levels over 14 days. For adults in their 50s and 60s where baseline NAD+ is significantly depleted, higher doses are needed to produce the same relative increase as lower doses achieve in younger adults.

A 500mg dual capsule typically contains 250mg NMN + 250mg Resveratrol — meaning your actual NMN intake is 250mg, not 500mg. This is a very common source of underdosing in this age group.
For adults over 60, the NAD+ deficit is at its most pronounced and higher doses are most warranted by the biology. Adults over 50: starting at 250–500mg/day and evaluating over 8+ weeks is a reasonable approach — consult with your physician, particularly if you take prescription medications.
